The kit is used for qualitative detection of cocaine content in human hair in vitro. Only used for the initial screening test of cocaine. For further confirmation or to obtain the exact concentration of the sample, more sensitive and specific detection methods are required, such as gas chromatography/mass spectrometry (GC/MS), high performance liquid chromatography (HPLC), etc.
The cocaine (COC) detection kit uses immunochromatography technology and uses the principle of competitive elimination to quantitatively detect the cocaine (COC) concentration in human hair samples. Antigen is fixed in the detection area (T area) of the nitrocellulose membrane in advance; after drug-specific monoclonal antibodies are labeled with fluorescent microspheres, they are fixed on the glass cellulose membrane to prepare a detection reagent strip. During the test, the hair is dissolved in a buffer solution, and after the sample is added, the liquid is chromatographed upward under the capillary effect. The fluorescent substance emits a fluorescent signal of a specific wavelength. Under the action of an excitation light source, the fluorescent analysis detector captures the excited fluorescent signal in the detection area (T zone), and the conversion of the fluorescent signal and the set calibration curve are automatically converted into quantitative values. Thus, the concentration of cocaine (COC) in the sample is calculated to realize the quantitative detection of the content of cocaine (COC) in the hair.

Testing method:
You must read the instruction manual completely before testing, and return the reagents and samples to room temperature (18-30°C) before use.
1. Prepare hair: Take 5mg of hair and cut it into 2-5mm length.
2. Unpack the aluminum foil bag of the test card and place the test card on a flat table.
3. a. Hair: Place the shredded hair in a centrifuge tube filled with 500μL of sample buffer and let it stand for 5-10 minutes.
b. Saliva: Take 50μL of the saliva sample and put it in a centrifuge tube containing 500μL of sample buffer, cover it and mix upside down and mix 5 times.
c. Urine and blood: Take 5 μL of urine and blood samples and put them into a centrifuge tube containing 500μL of sample buffer. Put the lid upside down and mix upside down for 5 times.
d. Dried blood spot: Take 5mg of dried blood spot, put it in a centrifuge tube filled with 500μL sample buffer, and let stand for 5-10min. Take 80μL of the above-prepared sample solution, add it to the sample card well, and let it stand at room temperature (18-30°C) for 5 minutes. Put the responding test card into the instrument to start measurement.
4. The hair drug trace detector matched with the kit has the function of calculating concentration. After the measurement is completed, the instrument automatically displays the concentration value on the screen.
Result judgment:
According to the Public Ban Drugs [2018] No. 930 "Drug-related Persons Hair Sample Testing Specification", the cutoff value of cocaine is 0.5ng/mg.
