Standard procedure for taking blood from laboratory animals
(A) Rabbits
- The method of collecting blood samples in the ear edge vein
Firstly, selected the ear vein and then removed the hair, clamping the root of the ear using a small vascular clip what could make venous dilatation. The experimenters held a thick needle from blood vessel of the tip ear into the vein to take blood, along the direction of the reverse flow, or cut veins with a scalpel, then blood flowed out. Collected 2-3 ml blood samples and pressed the wound with cotton balls to stop the bleeding. By pressing the collateral vein, the blood could flow out faster. Before collected the blood samples, rubbing liquid paraffin on the ear, what could prevent blood coagulation.
- The method of collecting blood sampling from the central ear artery
Rabbit was fixed in the box, rubbing the ear by hand, so that the central artery expansion. After that, Left hand clutched on the rabbit ears, right hand held on the syringe, then inserted the needle from the end of the central artery and pierced the artery along the direction paralleled to the artery. After collected 15 ml blood samples, pressed the cotton ball on the wound to stop bleeding. Note that the central arteries of the rabbit ears were prone to spasmodic contraction, so artery should be fully expanded before blood collection. It was more difficult to collect samples from thicker ear than the central artery.
- Collecting blood samples in the subcutaneous veins of hind limb tibia
Rabbit was fixed on the test bench, removed the hair of shin. Then the operator could tie a tourniquet on the thigh to make the subcutaneous vein of the lateral tibia expansion. Left hand pressed on the vein, right hand held syringes, keeping needle paralleled to venous blood vessels, subsequently the blood entered into the syringe when pulling back the Syringe plug.
(B) Guinea pigs
- The method of blood sampling from heart
Taking blood from the guinea pig's heart was basically the same as taking blood from a rabbit. The amount of blood could be determined according to the specific needs of the experiment, For example, some of the experiments needed blood 5-7ml, some needed 15-20ml.
- Foot vein blood sampling
The assistant fixed guinea pig with hands, straightening hind knee joint. Experimenter could found the back foot vein after routine disinfection. Left hand pulled the toe of guinea pig, simultaneously, right hand held the syringe needle. Then stop bleeding with gauze or cotton ball immediately when the samples collection was complete. Two hind legs could be alternately used while the experiment repeatedly needed blood collection.
(C) Dog
- The blood sampling method from heart
The Operation process of sampling blood from dogs was basically same to rabbit, except more blood can be sampled.
- The blood sampling method of venae saphena parva and cephalic vein
The direction of venae saphena parva extended from the lateral malleolus of hind limb to the backward side. The cephalic vein was located on the dorsal front of the forearm paw. Then cut off the local dorsal hair before taking blood, the assistant clenched legs of animals to make the subcutaneous veins filled, and the experimenter could draw blood by routine puncture smoothly.
